제목 Efficient Intracellular Translocation of Reductively Dissociable Dimeric siRNA Polyplexes for Targeted Gene Silencing
초록 The high stiffness and low spatial charge density of conventional siRNA limits its effective condensation with cationic polyelectrolytes. Here we report the effect of siRNA dimerization by hydrophobic spacers with three different lengths (three, six, and twelve carbons) on in vitro cellular uptake and gene silencing. Monomeric and dimeric siRNA molecules are electrostatically complexed with linear polyethylenimine (LPEI). All of the dimeric siRNA/LPEI complexes exhibit highly enhanced intracellular translocation and gene silencing effects compared to monomeric siRNA/LPEI complexes. It seems likely that the higher spatial charge density and increased chain flexibility of the siRNA dimers lead to the formation of more compact and stable siRNA/LPEI complexes.
