제목 Reducible Dextran-siRNA Conjugates for Stabilization, Tumor-targeting, and Efficient Gene Silencing
초록 Small interfering RNA (siRNA) has been considered as a very attractive therapeutic alternative to chemical-based medicines; however, their chemical and biological instability and poor delivery efficiency often limit the wide use for clinical applications. Here we report a self-assembled polyelectrolyte complex based on dextran-siRNA conjugates linked via reductively cleavable disulfide bonds. The conjugates were ionically complexed with linear polyethylenimine to a colloidal nanoparticle of about 200 nm in diameter. Dextran-siRNA conjugates provide effective structural features for stabilization and intracellular delivery of siRNA therapeutics. We expect that DSC can further improve siRNA delivery system not only by cationic polymeric cargos but by various nanoparticulate carriers.
