제목 Cartilage regeneration using in situ cross-linkable gelatin based hydrogels with primary chondrocytes
초록 In situ cross-linkable hydrogels have been widely used as a cell supporting scaffold for cartilage regeneration. In this study, in situ forming gelatin based hydrogels were developed as an injectable matrix for cartilage regeneration. The hydrogels were rapidly formed using horseradish peroxidase and hydrogen peroxide. Their physico-chemical properties could be controlled by varying the catalysts concentration. In vitro cell study was investigated using primary chondrocytes. The cells were cultured within the hydrogels formed with the different matrix stiffness and the cultured cells were evaluated by live/dead assay and RT-PCR. The cells were well proliferated with the hydrogels for 14 days. In addition, animal study was carried out using the rabbit chondral defects model. Obtained results suggested that in situ gelatin based hydrogels has a potential for cartilage regeneration and it can be also useful as an injectable material for biomedical applications.
