제목 Adhesion of vascular endothelial cells to polystyrene surface modified by active oxygen
초록 Polystyrene (PS) dish is widely used in the fields of tissue engineering and regenerative medicine for a cell culture substrate. Generally, plasma treatment is employed for enhancement of cell adhesion at the PS dish surface, and sterilization treatment of the PS dish is performed after the plasma treatment by gamma ray irradiation. Simultaneous process of the surface modification and sterilization is required to achieve simply and at a low-cost process. Active oxygen spices (AOS) has strong oxidation abilities and it is easy to generate from oxygen due to ultraviolet (UV) light irradiation. We have investigated surface modification and sterilization effects on the PS dish in a sterilization bag by using AOS generated via UV lamps. Furthermore, we have reported on the cell adhesiveness of osteoblast-like cell MC3T3-E1 on PS surface modified by active oxygen species. In this study, vascular endothelial cells HUVEC, which is poor adherent cell compared to MC3T3-E1, were cultured on the PS surface subjected to surface modification by active oxygen.  
Multi well PS dish wrapped into sterilization bag was used as a sample. The AOS exposure was carried out in a chamber equipped with a low-pressure mercury UV lamp, whose wavelength is 185 nm and 254 nm, to generate the AOS. After evacuating the chamber with a rotary pomp, oxygen gas was introduced up to atmospheric pressure. After exposure, chemical composition, surface roughness and wettability were analyzed by x-ray photoelectron spectroscopy (XPS), surface roughness analysis and water contact angles, respectively. And vascular endothelial cells (HUVEC) were cultured on the PS dish before and after AOS exposure at 37°C for 24 h in a 5% carbon dioxide incubator.  
The oxygen content of the PS surface was increased, and peak intensity represent functional group was increased after the AOS exposure. Water contact angle was decreased with increasing AOS exposure time. Relationship between contact angle and wettability of PS surface modified by active oxygen were confirmed. On untreated PS surface, spherical cells were observed, and the adhesion of the cells was poor. Wide spread shape cells can be observed on the PS dish after AOS exposure.  
Cell adhesion was also improved on PS dish due to the AOS exposure. The AOS treatment is one of effective methods to enhance the vascular endothelial cells adhesion.
