제목 Improved expression of cell penetrating scFv using an engineered ubiquitin in a cell-free protein synthesis system
초록 Antibody is in the core of current research for novel therapeutic and diagnostic agents. In particular, antibodies that can cross cellular membranes have potential applications in controlling disease-related intracellular protein-protein interactions. In this study, we developed an engineered ubiquitin sequence that substantially improves the expression level of cytosol-penetrating antibodies in a cell-free protein synthesis system. The initial codons of a ubiquitin gene was randomized, and the expression level of scFv fused with each of the variant ubiquitin sequences was examined to identify the optimal initial codons. As a result, we were able to select a number of variant ubiquitin clones that significantly improves the expression level of the fused cytosol-penetrating antibodies.
