제목 Injectable Extracellular Matrix (ECM) hydrogel derived from porcine skin loaded with bi-phasic calcium phosphate nano powders as a potential filler for bone regeneration
초록 Extracellular matrix is composed of abundant types of collagen, glycosaminoglycans and glycoproteins that makes it highly desirable for tissue engineering application.  In this study, abundant and easy to procure porcine skin was utilized to fabricate an injectable ECM derived hydrogel loaded with bi-phasic calcium phosphate powders (5% and 10% w/v) for bone regeneration. Porcine skin was decellularized using 2 different protocols, decellularization A contained 0.1% sodium dodecyl sulfate (SDS) in deionized water while decellularization B contained 0.1% sodium dodecyl sulfate (SDS) in 70% isopropanol. After decellularization, DNA content of the native and decellularized ECM were quantified and further confirmed by Hematoxylin and Eosin (H&E) staining and Hoechst staining while Oil Red O staining determined the delipidization process effectivity. Decellularized ECM content was assessed by quantifying some of the important components of the ECM such as collagen, sulfated glycosaminoglycan and elastin with native porcine skin as control. Injectable hydrogels loaded with BCP powders were assessed by Fourier transform infrared spectroscopy (FTIR), Energy dispersive X-ray spectroscopy (EDS), Scanning electron microscopy (SEM) and porosity analysis. Biocompatibility was determined using Bone marrow mesenchymal stem cells (BMSC) and in vivo studies were carried out by implanting injectable decellularized ECM hydrogels in rabbit’s  femoral head defect for 4 and 8 weeks to determine the effect of the decellularized ECM porcine skin derived injectable hydrogel on bone regeneration.
