제목 A Novel Strategy for Cancer Immunotherapy by Foreignizing Tumor cells Using a Polymeric Conjugate
초록 Cytotoxic T lymphocytes (CTLs) play important roles in immunological rejection in cancer immunotherapy. However, most tumors are not rejected by the immune system because they have self-antigens, which are not recognized by CTLs. In this study, we hypothesized that “foreignizing tumor cells” by delivering foreign antigen into the tumor cells might result in immunological rejection by CTLs. We prepared a polymeric conjugate including ovalbumin (OVA) as a foreign antigen and hyaluronic acid as a tumor-targeting ligand. It was effectively taken up by TC-1 cells and caused high OVA peptide presentation on the cellular surface. Furthermore, it was effectively accumulated into tumor site after I.V. injection, followed by substantial inhibition of tumor growth in mice model which is immunized with a vaccinia virus expressing OVA. These results demonstrated that the polymeric conjugates bearing foreign antigens are promising cancer immunotherapeutic agents by foreignizing tumor cells.
