제목 Effect of dexamethasone and ipriflavone on osteogenic differentiation of muscle-derived stem cells
초록 Abstract : Ipriflavone, a non-hormonal isoflavone derivative, has been shown to interfere with bone remodeling by inhibiting bone resorption and stimulating bone formation. Muscle-derived stem cells (MDSCs) can be differentiated in culture into osteoblasts, chondrocytes, and even myoblasts. In this study examines and compares the effects of dexamethsone and ipriflavone-dependent in vitro osteogenic differentiation of muscle-dericed stem cells. For osteogenic differentiation, MDSCs were cultured with osteogenic medium included 10-8M dexamethasone, DMEM, 10% FBS, 10-8M vitamin D3, 10mM β-glycerophospahte, 50㎍/㎖ L-ascorbic acid, and 10-8M ipriflavone, DMEM, 10% FBS, 10-8M vitamin D3, 10mM β-glycerophospahte, 50㎍/㎖ L-ascorbic acid. Cellular viability and proliferation were assayed by MTT test, and osteogenic differential cell were analyzed by alkaline phosphate activity, histochemical staining, and RT-PCR. We can be observed cell proliferation of MDSCs increased according to the time and ALP level which ipriflavone group is higher than dexamethasone group. In von Kossa staining, we can observed that ipriflavone group is higher than dexamethasone group by observing calcification.
