Preparation of itraconazole nanoparticles by anti-solvent precipitation method using a cascaded microfluidic device and an ultrasonic spray drier
We propose a cascaded device connecting microfluidic nanoprecipitation and ultrasonic spray drying to produce dry drug nanoparticles directly. Itraconazole (ITZ) was chosen as a model drug. Firstly, the influences of different factors such as the Reynold number, residence time, and volume flow ratio of solvent and anti-solvent on the particle preparation were investigated in a micromixer. Then, the micromixer was connected with an ultrasonic spray drier operated at tuned conditions so that dry ITZ nanoparticles with controllable particle size were prepared. Furthermore, ITZ nanoparticles with amorphous structure were obtained while using poly(vinylpyrrolidone- vinyl acetate) (PVPVA) as polymer stabilizer.
