제목 Amphiphilic Poly(Amino Acid) based Vaccine Adjuvants including Squalene for Influenza Virus Vaccine
초록 Depending on the outbreak of infectious diseases every year, the development of an anti-infectious disease vaccine is essential. However, the research of modern vaccine adjuvants for enhancing immunogenicity was slow in progress and types of commonly used adjuvants were limited. Here, we address amphiphilic graft poly(amino acid) (PLL-g-Phe) based nanoparticles including squalene (AASq) as a potent vaccine adjuvant against influenza virus. The results showed that the synthesized PLL-g-Phe loaded squalene (5% v/v) with proper loading efficiency and stability. More importantly, intramuscular co-administration of AASq(5%) and an H1N1 pandemic strain (CA04) produced antibodies and cytokines in a conditional mouse model. Furthermore, The AASq(5%)/CA04 immunization prevented disease symptoms and protected mouse model against lethal infection with H1N1 influenza virus. Consequently, our data indicate that AASq(5%) can be a great vaccine adjuvant for enhancing vaccine-induced immune responses
